[Review] Distinct neurological disorders with ATP1A3 mutations

Genetic research has shown that mutations that modify the protein-coding sequence of ATP1A3, the gene encoding the α3 subunit of Na+/K+-ATPase, cause both rapid-onset dystonia parkinsonism and alternating hemiplegia of childhood. These discoveries link ... [Published The Lancet Neurology - Apr 15 2014]

In vivo inhibition of the mitochondrial HATP synthase in neurons promotes metabolic preconditioning

A key transducer in energy conservation and signaling cell death is the mitochondrial H+-ATP synthase. The expression of the ATPase inhibitory factor 1 (IF1) is a strategy used by cancer cells to inhibit the activity of the H+-ATP synthase to generat ... [Published BioPortfolio - Apr 01 2014]

Quotes

According to the "ACC/AHA 2005 Guideline Update for the Diagnosis and Management of Chronic Heart Failure in the Adult" , S: Hunt et al., the disease continuum in the area of heart failure is nowadays grouped into four stages as noted above...
Our news editors obtained a quote from the research from Comenius University, "The aim of present study was the investigation of functional properties of cardiac Na, K-ATPase in 20-week-old male rats 6 weeks after c-irradiation by a dose 25 Gy (IR). Irradiation induced decrease of systolic blood pressure from 133 in controls to 85 mmHg in IR group together with hypertrophy of right ventricle (RV) and hypotrophy of left ventricle (LV). When activating the cardiac Na, K-ATPase with substrate, its activity was lower in IR in the whole concentration range of ATP. Evaluation of kinetic parameters revealed a decrease of the maximum velocity (Vmax) by 40 % with no changes in the value of Michaelis-Menten constant (Km). During activation with Na+, we observed a decrease of the enzyme activity in hearts from IR at all tested Na+ concentrations."
According to the news reporters, the research concluded: "Our findings suggest that BRCA2-dependent IIC-ring formation is a critical step in proper formation of the midbody, offering an explanation for how chromosome instability may arise in breast cancer."
"In the four stage ACC/AHA scheme, Stage A is asymptomatic but is at risk for developing HF. Stage B there is evidence of cardiac dysfunction without symptoms. In Stage C there is evidence of cardiac dysfunction with symptoms. In Stage D, the subject has symptoms of HF despite maximal therapy."
